Father Chris Riley's Op Shop has been bringing smiles to many in Bowral since 2002, and has been a fixture in Bundaroo Street for more than 16 years.
The premises was recently sold.
Op shop volunteer joint coordinators Susie Burnard and Sassi Cordony hope that the store can thrive in a new location, potentially in one of the region's town centres.
The store is one of three across NSW, whose funds go to Father Chris Riley's Youth Off the Streets programs.
Susie said the space gave the local community the opportunity to access quality household goods and clothing "for less".
"We perform a recycling service for the community too by accepting donations," Susie said.

"We are also catering to people who enjoy op shopping, it's part of the Highlands attraction.
"People know us and drop in regularly to see what we've got."
Sassi said there has been an increase in "very high quality, fabulous labels" that were not there before.
Customers can discover items such as bric-a-brac, kitchenware, books and clothing at bargain prices.
The pair, and other volunteers, would love to offer the same services in a new location, and said wheelchair accessibility, parking spaces and an area for a skit bin were important.
Sassi said having space where people could drop off donations, and room for shelves and storage were also beneficial.
If anyone can help, or has any information about a new home for the op shop, they can call Susie on 0406488262 or email psburnard@gmail.com.
Father Chris Riley's Op Shop Bowral is open from 9.30am to 4pm Monday to Friday and 9.30am to 1pm on Saturday.
