The centre has been temporarily closed and is undergoing deep cleaning, but will re-open on Monday, September 20.
The call for testing covers about 200 patients who were at the centre last week between Monday, August 30 and Friday, September 3, and who are considered close contacts.
Centre staff are reaching out to as many patients as possible and in the interim have put out a public call for patients to get tested.
Regal Medical Centre said the outcome was a timely reminder for people to adhere to COVID-19 protocols, including social distancing, isolating and mask wearing.
People with COVID-19 symptoms including coughing and sneezing are encouraged to visit local testing centres.
There are two testing locations in the Southern Highlands.
Bowral and District Hospital. Open 8:30am to 5pm, seven days a week. Appointments not required. Entry is from Ascot Road.
Mittagong Respiratory Clinic. Open 8am to 5om, seven days a week. Appointments required, call 4800 9032. The clinic is located in the lower level of Mittagong Health Care Centre.
