More than 3000 riders are registered to participate in the event.
Bowral Classic organiser, James Yaffa said that riders should remain off the roads where possible to protect the safety of the cyclists.
Bong Bong Street will be closed from 4am until 9am for the ride.
Range Road, Tourist Road and Eridge Park Road will also be closed to traffic except local residents from 9am to 3pm.
